Leu-enkephalin Aquamax-aequorin fusion protein

 

Application/Background 

Opioid peptides are involved in a series of physiological processes such as analgesia, stress, immune response, blood pressure regulation, tumor growth, memory and regulation of food consumption. The smallest of these peptides are the pentapeptides methionine- and leucine-enkephalin and they differ only in their C-terminal amino acid.  These neuropeptides mediate the control of numerous physiological systems including the cardiovascular, autonomic, endocrine, and immune systems. Elevated levels of these neuropeptides are found in patients with Down syndrome and Alzheimer’s disease.  Aquamax-Leu-enkephalin has been used in the development of sensitive assays for Leu-enkephalin (1).

 

Dose-response curve for Leu-enkephalin generated in a competitive binding mode. A limit of detection of 1 x 10-9 M was achieved.

 

 

Dose-response curve for Leu-enkephalin generated in sequential binding mode.  This assay produced a limit of detection of 3 x 10-10 M.  

Both of the assays produced a working range of 3 orders of magnitude (1). (This is different from the dynamic range presented in the table from the SenseOmics presentation which shows a dynamic range of  10-4 to 10-10. )
